The eligibility criteria for RTO posts differ as there are 3 different posts involved in this. You are eligible to write RTO clerkship posts after 12th class or diploma. A B.E/B.Tech degree graduate can apply for RTO Assistant engineering position. On the other hand, an LLB graduate can apply for RTO Judicial post. As part of selection process for any of these posts a written exam need to be attended. You will have to study subject areas like General studies, optional subjects, language skills etc. You will have to attend PSC exam of the concerned state from which you are applying to get in to any of the RTO posts.
