I'm doing my 12th from RIOS Board (Rural Institute of Open Schooling). Can I get admission in regular colleges or not for pursuing B.Com or any other graduate level courses from a regular college/university?

Definitely you can take admission in regular college after passing in 12th standard from RIOS board.
RIOS stands for rural Institute of Open Schooling. It is a valid board like any other state board.

One who has completed intermediate from RIOS board is definitely eligible for whatever course or job he or she wants to apply.
This certificate is a valid certificate and recognised by all the universities in our country.
