I have completed my 3 years Diploma course as a regular candidate. Now, can I pursue Diploma in Education (D.Ed.) and B.Ed thereafter? Or, is graduation compulsory for D.Ed and B.Ed?

D.Ed refers to Diploma in Education and to pursue the same course you are required to have completed your higher secondary school degree i.e; Class 12th in Science or Humanities stream from a recognized board of education.

So, having completed 3 year Diploma course, you are eligible for D.Ed. only if you have pursued a regular higher secondary school degree course too otherwise not.

Also, I would like to let you know is that for pursuing B.Ed ( Bachelor of Education ) degree course, you are required to hold a bachelor degree course with at least 50% marks and therefore to pursue B.Ed you will have to first pursue a graduation degree course too as you cannot opt for the same degree course on the basis of D.Ed degree course.
