The National Institute of Virology was established in 1952 as Virus Research Centre (VRC). It is one of the major Institutes of the Indian Council of Medical Research (ICMR). It became one of the collaborating laboratories of the World Health Organization (WHO) in 1967. It became a regional centre of the WHO for South-East Asia for arbovirus studies from the year 1969.

The institute began its function in the year 1974 as a collaborating center for the World Health Organization for arbovirus research and reference. It was re-designated as the WHO Collaborating Centre for Arbovirus and Haemorrhagic Fever Reference and Research and Rapid Diagnosis of Viral Diseases in 1995. In the year 1997, the institute became a part of the Global Polio Eradication Programme of the World Health Organization in the region of South-East Asia.
