AUCET Applied Biochemistry Entrance Test 2012

Structure of the AUCET Applied Biochemistry Entrance Test
Comprising of 90 multiple choice objective type questions, the question paper will have 3 sections. Section A, B, and C with 40, 30 and 20 questions respectively. Each section carries 1 mark for each question. ¼ marks will be deducted for each incorrect answer. Duration of the entrance test is 75 minutes .

Molecules And Their Interaction Relevant To Biology: Structure, function and composition of biomolecules (carbohydrates, lipids, proteins, nucleic acids and vitamins), Biophysical chemistry principles (pH, buffer, reaction kinetics, thermodynamics, colligative properties), Enzymes and enzyme kinetics, mechanism of enzyme catalysis, enzyme regulation, Principles of catalysis and isozymes, Glycolysis, Coupled reaction, Oxidative phosphorylation, Group transfer, Bioenergetics, Biological energy transducers, Conformation of nucleic acids (A-, B-, Z-, DNA), t-RNA, micro-RNA): Conformation of proteins (Ramachandran plot, secondary, tertiary and quaternary structure; domains; motif and folds), Enzymes and enzyme kinetics
Cellular Organization: Membrane structure and function: Structure of model membrane, Osmosis Ion channels, Lipid bilayer and membrane protein diffusion, Ion pumps Mechanism of sorting and regulation of intracellular transport, Active transport Electrical properties of membranes
Structural organization and function of intracellular organelles: Nucleus, lysosomes, mitochondria, Golgi bodies, Cell wall, endoplasmic reticulum, peroxisomes, vacuoles, chloroplast, structure & function of cytoskeleton and its role in motility, plastids
Organization of genes and chromosomes: Gene families, structure of chromatin and chromosomes, interrupted genes, unique and repetitive DNA, transposons, heterochromatin, and euchromatin
Fundamental Processes DNA replication, repair and recombination: Unit of replication, replication origin and replication fork, enzymes involved fidelity of replication, DNA damage and repair mechanisms, extra-chromosomal replicons
RNA synthesis and processing: Formation of initiation complex, transcription repressors and activators, Transcription factors and machinery, RNA polymerases, elongation and termination, capping, RNA processing, splicing, RNA editing, RNA transport, polyadenylation, structure and function of various types of RNA
Protein synthesis and processing: Formation of initiation complex, initiation factors - Definition and their regulation, Ribosome, elongation and elongation factors, genetic code, termination, tRNA-identity, aminoacylation of tRNA, aminoacyl tRNA synthetase, translational inhibitors, posttranslational modification of proteins, translational proof-reading
Control of gene expression at transcription and translation level: Viruses, Regulation of phages, role of chromatin in regulating gene expression and gene silencing, prokaryotic and eukaryotic gene expression

Structure and function of antibody molecules, B and T cell epitopes, generation of antibody diversity, antibody engineering, monoclonal antibodies, antigen - antibody interactions, antigen processing and presentation, MHC molecules, activation and differentiation of B and T cells, humoral and cell-mediated immune responses, primary and secondary immune modulation, parasitic (malaria) and viral (HIV) infections, congenital and acquired immunodeficiencies, B and T cell receptors, the complement system, cell-mediated effector functions, inflammation, hypersensitivity and autoimmunity, Toll-like receptors, immune response during bacterial (tuberculosis), vaccines
Applied Biology: Application of immunological principles (vaccines, diagnostics), Microbial fermentation and production of small and macro molecules, tissue and cell culture methods for plants and animals, Genomics and its application to health and agriculture, including gene therapy, Bioremediation, Transgenic animals and plants, molecular approaches to diagnosis and strain identification, Biosensors, Plasma proteins and their variations in diseases, Anemia, Clinical Enzymology, Liver and Kidney Function tests, Endocrine disorders of Pancreas
Methods In Biology - Molecular biology and recombinant DNA methods: DNA (genomic and plasmid) and proteins, Isolation and purification of RNA, different separation methods, DNA and proteins by one and two dimensional gel electrophoresis, analysis of RNA, isoelectric focusing gels, expression of recombinant proteins using bacterial, animal and plant vectors, molecular cloning of DNA or RNA fragments in bacterial and eukaryotic systems, generation of genomic and CDNA libraries in plasmid, BAC and YAC vectors, phage, cosmid, in vitro mutagenesis and deletion techniques, gene knock out in bacterial and eukaryotic organisms, protein sequencing methods, detection of post-translation modification of proteins, RFLP, RAPD and AFLP techniques. DNA sequencing methods, strategies for genome sequencing, isolation of specific nucleic acid sequences, methods for analysis of gene expression at RNA and protein level, large scale expression analysis, such as micro array based techniques, isolation, separation and analysis of carbohydrate and lipid molecules
Histochemical and immunotechniques: Detection of molecules using ELISA, RIA, Antibody generation, western blot, flowcytometry and immunofluorescence microscopy, immunoprecipitation, detection of molecules in living cells, in situ localization by techniques such as FISH and GISH
Statistical Methods: T-test, analysis of variance test. Measures of central tendency and dispersal, probability distributions (Binomial, Poisson and normal), difference between parametric and non-parametric statistics, confidence interval, errors, levels of significance, sampling distribution, regression and correlation
Biophysical methods: Circular dichroism, NMR and ESR spectroscopy, Analysis of biomolecules using UV/visible, fluorescence, structure determination using X-ray diffraction and NMR, analysis using light scattering, different types of mass spectrometry and surface plasma resonance methods
Microscopic techniques: Visualization of cells and sub-cellular components by light microscopy, resolving powers of different microscopes, microscopy of living cells, scanning and transmission microscopes, different fixation and staining techniques for EM, freeze-etch and freeze-fracture methods for EM, image processing methods in microscopy
Radiolabeling techniques: Properties of different types of radioisotopes normally used in biology, Radioisotopes detection and measurement, radioisotopes incorporation in biological cells and tissues, safety guidelines molecular imaging of radioactive material
Computational methods: Data mining methods for sequence analysis, web-based tools for sequence searches, motif analysis and presentation, Nucleic acid and protein sequence databases
Eligibility Criteria
Qualified candidates with a B.Sc degree from any recognized university for any of the following disciplines are considered eligible and final year B.Sc students for the same below can apply.

How to Apply?
A payment of Rs 300/- can get you an application form for this exam from authorized sales counters only, which will be duly informed. To receive the form by post, a DD of Rs 350/- drawn in favor of ‘The Director’, AUCET, Directorate of Admissions should be sent along with a request. The DD should be payable at Vishakapatnam only. After filling up the form, send it to the director as early as possible.
How to fill the Form?
A neatly and precisely filled application form is recommended from candidates. All entries must be correct. Candidates must take care not to make any errors and hence a double check after filling up the form is advised. Entries in the forms should be made the applicant itself in their own handwriting.
